Summary

THE HEARTS WERE PERFUSED WITH 5-10 ML OF CRYOPROTECTANTS: GLYCEROL, ETHYLENE GLYCOL AND DMSO IN RINGER'S SOLUTION BY THE PRESSURE OF 13.33 KPASCAL. THE VARIABLES INCLUDED FIVE CONCENTRATIONS: 1, 2, 3, 4, AND 5 MOL. THE EXPERIMENTS INCLUDED THE PRESERVATION OF WHOLE ORGANS AND TRIMMED PIECES OF THE HEART WHICH WERE WRAPPED IN ALUMINIUM FOIL AND COOLED AT 1 K/MIN TO 193 K (-80 DEG C), AFTER WHICH THEY WERE MAINTAINED IN LIQUID NITROGEN FOR 42 DAYS, ONE AND TWO YEARS. THE SAMPLES WERE THAWED BY ABRUPT IMMERSION INTO RINGER'S SOLUTION AT 311 K (38 DEG C). THE RESUMPTION OF ACTIVITY WAS EXAMINED UNDER THE MICROSCOPE AND RECORDED BY A MOVIE CAMERA. A PACE-MAKER WAS USED TO STIMULATE THE SAMPLES. THE MAXIMUM LENGTH OF PRESERVATION IN LIQUID NITROGEN WAS SIX YEARS.
