Summary

A low toxicity vitrification solution, designated EFS40, was composed of representatives of three different categories of agents, that is, ethylene glycol as a rapidly permeating low toxic agent, Ficoll 70 as a macro-molecule, and sucrose as a small saccharide. This solution is efficient for vitrifying mouse morulae after 0.5-5 min of exposure at 20 deg C. However the optimum time of exposure will depend on the temperature and on the developmental stage and the species of embryos. If embryos are likely to be injured before sufficient permeation of the cryoprotectant, a two-step method improves the survival. Fracture damage can be prevented by slightly low cooling and warming. In mouse embryos, permeated ethylene glycol can be removed efficiently in a sucrose solution. Vitrification of embryos using EFS40 will also be available for embryos of other species.
