Summary

THE PRINCIPLE OF THE TWO NEW METHODS IS THAT THE DAMAGE CAUSED TO THE LIVER MITOCHONDRIA BY FREEZING RELEASES THE MITOCHONDRIAL ENZYME BETA-HYDROXYACYL-COA-DEHYDROGENASE (HADH) INTO THE CELL PLASMA OF THE LIVER TISSUE. INCREASED HADH ACTIVITY INDICATES THAT THE LIVER HAS BEEN FROZEN AND THAWED. IN ONE METHOD THE RELEASED HADH ACTIVITY IS MEASURED PHOTOMETRICALLY AND IN THE OTHER IT IS DETERMINED BY MEANS OF A SIMPLE COLOUR REACTION. THE METHODS CAN BE USED ON LIVER TISSUE FROM CATTLE AND PIGS. THEIR RELIABILITY IS NOT IMPAIRED BY KEEPING THE LIVER CHILLED AT 275 K (2 DEG C) FOR SEVEN DAYS.
