Summary

Optimal pretreatments, freezing and postculture conditions were studied for grapevine shoot tips cryopreserved by the encapsulation-dehydration process. Best results (40-60% survival and 30% shoot recovery without callus formation) were obtained after a 2-day stepwise preculture in a medium containing 1 M sucrose, two-step cooling, and direct subculture on standard culture medium. Higher survival rates were noted at lower precooling temperatures and cooling rates. Tolerance of shoot tips was independent of their position on the stem.
